Aqueous hybridization- or detection buffer comprises at least a water-soluble source for unsubstituted ammonium ion, at least a buffer substance that stabilizes the pH of the buffer to 6.5-9, at least a chelating agent, at least a surfactant, and optionally at least a polymeric solubilizer. An independent claim is included for selective detection of target nucleotide sequences in a sample, comprising (a) hybridization of target nucleotide sequences contained in the sample with complementary oligonucleotide sequences (probes) under conditions which allows the hybridization of complementary nucleotide sequences to form hybrids, by mixing the target nucleotide sequences and the probes in the buffer solution, (b) conversion of the resulting hybrids with a detection reagent under the formation of labeled hybrids and (c) detection of the selected hybrids. |
