The present invention relates to a method for mass-propagating NK cells, comprising the steps of: isolating lymphocytes from the peripheral blood of a specific volume (30-100 ml) allowable by single blood collection; transferring the isolated lymphocytes into a flask, followed by primary culturing in the presence of a composition for culturing immune cells; collecting only NK cells from the cultured immune cells, followed by secondary culturing in a flask; collecting the NK cells after the secondary culturing, followed by tertiary culturing in a flask having a larger volume than the flasks for the previous culturing; and collecting the NK cells after the tertiary culturing, followed by quaternary culturing in a large-volume flask. According to the present invention, the culturing was performed through four times of culturing after the isolation of the NK cells, and thus the NK cells can be propagated by 1000 fold or higher based on the initial count of NK cells. Further, there can be provided a composition for preventing and treating cancers, containing the propagated and cultured NK cells as an active ingredient.
