PROBLEM TO BE SOLVED: To provide nucleotides having a removable protective group, their use in polynucleotide sequencing methods, and a method for chemical deprotection of the protective group.SOLUTION: The modified nucleotide or nucleoside molecule comprises a purine or a pyrimidine base and a ribose or a deoxyribose sugar moiety having a removable 3'-OH blocking group covalently attached thereto, such that the 3' carbon atom has attached a group of the structure -O-Z where Z is any of -C(R')2-O-R'', -C(R')2-N(R'')2, -C(R')2-N(H)R'', -C(R')2-S-R'' and -C(R')2-F, where R' is, for example, an alkyl or a substituted alkyl, Z is an azidomethyl, R'' is a benzyl or a substituted benzyl. |
