The present invention relates to: a protein biosensor comprising a pore structure using a diffusive flow which uses the pore structure for checking the existence of antigens and measuring the concentration of the antigens in target fluid using the diffusive flow without the external power supply; and a fabricating method thereof. The protein biosensor using a diffusive flow by the present invention comprises the following: a nanopore formed on the upper side of a substrate in a hollow hole form for supplying target fluid; a first micropore formed on both ends of the nanopore with a step, having a bigger diameter than the nanopore; and a second micropore formed on both ends of the first micropore with a step, having a bigger diameter than the first micropore. Standard fluid is supplied to the lower side of the substrate. |
