PROBLEM TO BE SOLVED: To provide a one step method for producing an antigen-loaded antigen-presenting cell from monocytes ex vivo.SOLUTION: An antigen-loaded antigen-presenting cell is produced ex vivo from monocytes by contacting TNFα and a granulocyte macrophage colony stimulating factor with monocytes to form the antigen-presenting cell at a first point of time, and by contacting a soluble or particulate antigenic substance with the antigen-presenting cell to form the antigen-loaded antigen-presenting cell at a second point of time. The antigen-presenting cell is composed of two or more subsets selected from a group consisting of a Langerhans cell having a surface marker (CD1a+ CD207+), an interstitial dendritic cell having a surface marker (CD1a+ CD207-), a double negative dendritic cell having a surface marker (CD1a-CD14-), and a dentritic cell having a surface marker (CD14+CD1a-CD209+). |
