PROBLEM TO BE SOLVED: To provide a technique to solve the problem that, for a specimen such as a biological molecule having relatively large electromagnetic wave absorption, it is difficult to calculate refractive index distribution or dielectric constant distribution in the depth direction of the sample.SOLUTION: Provided is a method of calculating a refractive index of at least a part of a specimen by using electromagnetic wave measurement, the method at least including: a step 100 of measuring a first scattered wave from a structure of the specimen; a step 110 of measuring a second scattered wave from a structure in which a material 111 for calculating refractive index is disposed on a surface of the specimen; and a step 122 of comparing intensities at corresponding portions of the first scattered wave and the second scattered wave.
