PURPOSE: A cryopreservation method of amniotic fluid-derived stem cells is provided to largely reduce the concentration of Me_2SO, to remove animal serum (fetal bovine serum, FBS), and to enable long-term cryopreservation of the stem cells without damage to the stem cells using trehalose, sucrose, and catalase. CONSTITUTION: A medium composition for cryopreservation of amniotic fluid-derived stem cells is prepared by adding 2.5-5 %(v/v) of Me_2SO and catalase to an FBS-free medium containing trehalose, catalase, and zVAD-fmk. The medium suppresses free radical generation and cell apoptosis and maintains a stem cell differentiation ability. An amniotic fluid-derived stem cell composition is stored in liquid nitrogen by mixing the medium composition and the amniotic fluid-derived stem cells. |
