The invention relates to high throughput assays to evaluate antibodies that bind to HIV-1. The invention relates to methods of detecting antibodies capable of inhibiting the binding of HIV gp120 to alpha4beta7 integrin. The methods comprise administering a candidate antibody to the alpha4beta7 integrin in the presence of a binding protein, and determining if the candidate antibody blocks the binding of the binding protein to the alpha4beta7 integrin. The lack of candidate antibody blocking the binding of the binding peptide to the alpha4beta7 integrin is indicative that the candidate antibody will not inhibit the binding of HIV gp120 to the alpha4beta7 integrin.
