发明名称 METHOD FOR INTRODUCING EXOGENOUS DNA BY OVERCOMING RESTRICTION MODIFICATION BARRIER OF TARGET BACTERIUM
摘要 Disclosed is a method for introducing an exogenous DNA by overcoming a restriction modification barrier of a target bacterium. The method provided by the present invention comprises the following steps: 1) co-expressing all DNA methyltransferase encoding genes of a genome of a target bacterium in an Escherichia coli with no self restriction modification system, so as to obtain a recombinant strain A; 2) introducing an exogenous DNA molecule into the recombinant strain A for internal modification, so as to obtain an exogenous DNA molecule modified by methylation; and 3) introducing the exogenous DNA molecule modified by the methylation into the target bacterium. The experiment of the present invention proves that compared with an existing method for overcoming a restriction modification barrier of a bacterium to implement a genetic operation, the method provided by the present invention achieves a higher conversion rate.
