The invention provides compositions and methods for reducing and preventing tumor escape from immunotherapy. Specifically, the invention relates to administering a genetically modified T cell comprising both a TCR and a CAR. The invention provides an isolated nucleic acid encoding a T cell receptor (TCR) and a chimeric antigen receptor (CAR), wherein the isolated nucleic acid sequence comprises a TCR-a sequence, a TCR-beta sequence, a CAR antigen binding domain and a costimulatory signaling domain sequence. In one embodiment, the CAR sequence further comprises the nucleic acid sequence of a CD3 zeta signaling domain.
