A method for determination of plant peroxidase enzymatic activity, comprising preparation of reaction mixture with using of substrate-reducer (3,3',5,5'- tetramethyl benzidine dihydrochloride monohydrate), substrate-oxidizer (hydrogen peroxide), enzymatic preparation, buffer solution, registering by spectrophotometrical method the values of optical density at wavelength 650 nm, determination of protein concentration in enzymatic preparation, calculation of peroxidase specific activity, wherein before preparation of reaction mixture the preparation of unpurified tissue enzymatic extracts of cytoplasmic and ion-joined peroxidase is carried out, which are used as enzymatic preparations; hydroperite is used for preparation of substrate-oxidizer, two citric-phosphatic buffer solutions are prepared with pH 4.8 with concentration of 0.05 and , and potassium chloride is additionally introduced into the second solution in amount of , wherein protein concentration is determined by Bredford method with Coomassi Briliant Blue G250. |
