The method involves producing a digital hologram in an off-axis manner from a selected region of a sample (1). A focus position to be adjusted is determined along an optical axis (z) of a microscope lens (10) based on a digital hologram. The selected region of the sample is arranged in focus in best possible way. The microscope is adjusted to the determined focus position by a control and focused on the selected region. Partial images are recorded in the determined and adjusted focus position. The partial images are combined to an image stack along the optical axis. An independent claim is also included for a microscope. |
