PURPOSE: A method for preparing a physiologically active polypeptide conjugate is provided to highly maintain activity in vivo, to remarkably increase a half life in blood, and to enhance drug compliance. CONSTITUTION: A method for preparing a physiologically active polypeptide and non-polypeptide polymer connector comprises: a step of reacting a physiologically active polypeptide and a non-polypeptide polymer in a reaction solution containing an organic solvent; and a step of isolating and purifying the physiologically active polypeptide connector from the reacted mixture. The organic solvent is C1-C10 alcohol. The alcohol is selected from the group consisting of isopropanol, ethanol, and methanol. The reaction solution contains 10-60wt% of organic solvent.
