<P>PROBLEM TO BE SOLVED: To provide a method for modifying amino acids in an antibody variable domain in order to efficiently purifying a bispecific antibody; a pharmaceutical composition composed of the modified bispecific antibody; and a method for producing the bispecific antibody pharmaceutical composition. <P>SOLUTION: There are disclosed a method for efficiently purifying the bispecific antibody with a chromatography column by introducing difference of isoelectric points between H-chains of two kinds of antibodies through modifying amino acids existing on the surface of antibody variable domains by the two kinds of antibodies composing the bispecific antibody, and utilizing the difference of isoelectric points; and a method for efficiently purifying the bispecific antibody with a chromatography column by incorporating each antigen-binding site (heavy-chain variable domain) into a constant domain of antibodies with difference of isoelectric points, and co-expressing these. <P>COPYRIGHT: (C)2013,JPO&INPIT |
