A hemodialysis system (10) includes a dialysis instrument (36), first and second peristaltic dialysate pumps (22) operated by the instrument, a cassette (40) including a rigid structure and first and second dialysate pumping tubes (44) carried by the rigid structure, and a mechanism (110) configured to (i) initially allow the cassette (40) to be accepted by the dialysis instrument and (ii) thereafter move a member downwardly onto the first and second dialysate pumping tubes for operation with the first and second peristaltic dialysate pumps.
