Immobilizing nucleic acid ligands including at least one reactive amine function, by grafting on an activated solid substrate, including a step of coupling the nucleic acids on the activated solid substrate having a pH of less than 6. Independent claims are included for: (1) immobilizing nucleic acid ligands comprising at least a primary amine function on a solid support, comprising providing a solid support on its surface of active carboxylic acid groups, providing a nucleic acid ligand comprising at least one primary amine function, and coupling the nucleic acid with active carboxylic acid groups present at the surface of the solid support under conditions of pH of less than 6; (2) preparing an affinity support, comprising implementing the immobilization process; (3) the solid support of affinity obtained by the preparation process; (4) a complex resulting from the interaction of a nucleic acid ligand and a target molecule, where the complex is formed on the surface of a solid support; and (5) purifying a target molecule with affinity support, comprising contacting a composition to be purified comprising the target molecule with a solid affinity support to form a complex between nucleic acid ligands grafted on the support and the target molecule, and releasing the target molecule from the complex and recovering the purified target molecule. |
