PURPOSE: A PAUF-specific aptamer and a composition containing the same for treating pancreatic cancer are provided to ensure high affinity and specificity without toxicity and immunogenicity. CONSTITUTION: A PAUF(pancreatic adenocarcinoma up-regulated factor)-specific aptamer P12FR2 has a base sequence of sequence number 4. The aptamer is deformed to have RNase resistance by: substituting -OH group at 2' carbon site of saccharide structure in one or more nucleotide into -Me(methyl), -OMe, -NH_2, -F(fluorine), -O-2-methoxyethyl -O-propyl, -O-2-methylthioethyl, -O-3-aminopropyl, -O-3-dimethyl aminopropyl, -O-N-methyl acetate amido, or -O-dimethyl amidooxyethyl; substituting oxygen in a saccharide structure in the nucleotide into sulfur; and phosphorothiate, borano phosphate, or methyl phosphonate bond. A composition for treating cancer contains PAUF-specific aptamer P12FR2. |
