[Problem] To provide: a two-dimensional cell spreading device which can separate a desired cell from a cell having a smaller diameter than that of the desired cell and can arrange the desired cell and a cell having a similar diameter to that of the desired cell densely in a planar state when a cell suspension comprising multiple types of cells is spread on a plane surface; and a cell spreading method using the two-dimensional cell spreading device. [Solution] A two-dimensional cell spreading device which can spread a desired cell contained in a cell suspension in a planar state and has at least one groove formed thereon, said two-dimensional cell spreading derive being characterized in that the largest width of the upper surface of an opening of the groove is equal to or larger than the diameter of the desired cell, the groove has a smaller width than the diameter of the desired cell in the depth direction so as to hold the desired cell in the middle of the depth direction, and the bottom of the groove has a slit having a width of 1 µm or less or a pore having a diameter of 1 µm or less or does not have the slit or the pore.
