PURPOSE: A method for preparing mature dendritic cells is provided to improve titer and to manufacture various dendritic cell vaccines. CONSTITUTION: A method for preparing mature dendritic cells comprises: a step of collecting immature dendritic cells; and a step of stimulating the immature dendritic cells by combination of cytokine cocktail and poly(I:C) to induce differentiation into mature dendritic cells. The cytokine cocktail contains IL-1β, IL-6, TNF-alph, PGE2 and poly(I:C). The cytokine cocktail is applied to the immature dendritic cells before application of poly(I:C). The immature dendritic cells are derived from monocytes and hematopoietic stem cell from peripheral blood, umbilical cord blood, or bone marrow. A method for manufacturing the vaccine comprises a step of co-culturing the mature dendritic cells and antigen. The dendritic cell vaccine is a cancer vaccine. |
