Disclosed are assays for determining the presence or absence of active botulinum toxin serotype A (BoNT/A) in a sample and for determining the activity/potency of a BoNT/A preparation. Further disclosed are compositions for producing alpha-SNAP-25 antibodies, comprising a carrier, a flexible linker and a SNAP-25 antigen. Also disclosed is an isolated alpha-SNAP-25 antibody that specifically binds to an epitope comprising a carboxyl-terminus glutamine from the BoNT/A cleavage site scissile bond from a SNAP-25 cleavage product.
