<P>PROBLEM TO BE SOLVED: To provide a method for acquiring and amplifying polynucleotide (tester-specific polynucleotide), in which the amount existing in a test sample (a tester) is larger than the amount existing in another test sample (a driver), efficiently, easily and in a short period of time. <P>SOLUTION: The method for amplifying the tester-specific polynucleotide includes: (1) a process of performing hybridization by mixing a single-stranded polynucleotide derived from the tester with a single-stranded polynucleotide derived from the driver which may be a complimentary strand with the single-stranded polynucleotide derived from the tester; (2) a process of removing a hybridized double-stranded polynucleotide by oxygen treatment; (3) a process of amplifying non-hybridized single-stranded polynucleotide derived from the tester; and (4) a process of removing the non-hybridized single-stranded polynucleotide derived from the driver in the process (1) by the oxygen treatment. <P>COPYRIGHT: (C)2012,JPO&INPIT |
