<P>PROBLEM TO BE SOLVED: To provide a column which can be manufactured with an axial compression packing method and components, and further, is small-sized and disposable. <P>SOLUTION: A slurry chamber is fastened to and held in coaxial alignment with a chromatography column 122a by a V-band. One end of the column 122a has an end cap 48 and flit 42. A slurry of fluid and chromatographic media is placed in the column 122a and chamber. A piston 40 is forced through the chamber and into the column 122a by a hydraulic ram to expel the fluid and form a media bed in the column 122a. The slurry chamber is removed, and a split end cap 48 is fastened to the column 122a. A threaded recess 135b in the split end cap 48 receives a split, threaded collet 144. The collet 144 is tightened until the collet 144 pushes against the piston 40 with the same force as the ram, at which point the ram is released and the column 122a is removed for use. <P>COPYRIGHT: (C)2012,JPO&INPIT |
