PURPOSE: A cultivation method of mushroom containing omega-3 using a fermentation container is provided to use a medium having high moisturizing capacity for fermenting. CONSTITUTION: A cultivation method of mushroom containing omega-3 using a fermentation container comprises the following steps: inserting materials into the fermentation container, and rotating for mixing(S2); generating ammonia emitting heat by decomposing carbohydrate and decomposing protein using microorganisms(S3); softening a mushroom culture medium using the heat increase caused by the ammonia(S4); cooling the softened mushroom culture medium(S5); inserting the cooled mushroom culture medium with mushroom hyphae into a plastic bag(S6); injecting the mushroom culture medium into mushroom fungi(S7); and forming primordial of the mushroom for culturing the mushroom, and inserting the medium into a bottle for inserting strains(S8).
