PROBLEM TO BE SOLVED: To provide a method of amplifying nucleic acid for obtaining a nearly constant amount of an amplification product regardless of the amount of RNA to be used as a template.

SOLUTION: In a reaction for synthesizing a single-stranded cDNA using an RNA extracted from a biological sample as a template and using a primer having an oligo dT and a promoter sequence, the method for synthesizing a single-stranded cDNA includes using the primer at a final concentration of 500 pM-500 nM.
