Modified release pharmaceutical formulations and methods for enhanced mucosal drug absorption. The formulation comprises initial population(s) of particles comprising both drug and penetration enhancer which are released at a first location in the gastrointestinal tract, and a subsequent population or populations of particles comprising a penetration enhancer(s) having a delayed release due to a polymeric coating or matrix. This penetration enhancer is released at an additional location(s) in the intestine downstream from the first location and enhances absorption of the drug when it reaches the additional location(s).
