A device (20) is disclosed for determining the presence of an analyte in a liquid sample. The device comprises a test strip (131) defining a flow path. The test strip (131) comprises a second reagent zone (201) positioned along the flow path, a first reagent zone (121) located downstream of the second reagent zone (201) and a test zone (132) located downstream of the first reagent zone (121). The second reagent zone (201) comprises a first binding moiety being mobilizable by the liquid sample, the first binding moiety being capable of binding with the analyte to form a first complex. The first reagent zone (121) comprises a second binding moiety being mobilizable by the liquid sample, the second binding moiety being capable of binding with the first complex to form a second complex and comprising a detectable label. The test zone (132) comprises a third binding moiety being immobilized in the test zone (132), the third binding moiety capable of binding with the first or second complex. The second reagent zone (201) is separable from the first reagent zone (121) and prior to placing the second reagent zone (201) in fluid communication with the test strip (131), the liquid sample has already been in contact with the second reagent zone (201) for a period of time sufficient to effect formation of the first complex.
