<P>PROBLEM TO BE SOLVED: To provide a method for detecting, in a highly sensible and specific manner, an autoantibody in serum such as an HCC patient specific autoantibody from the serum of an HCV infected person. <P>SOLUTION: The present invention relates to an autoantibody detection method including the steps of: preparing a conjugated protein containing an antigen protein, GFP united to an N terminal of the antigen protein, and a cysteine tag united to a C terminal; preparing an antigen protein chip by reacting the cysteine tag of the conjugated protein and a maleimide group on a substrate surface to which the maleimide group is introduced via an amino group to fix the conjugated protein on the substrate surface through covalent bonding; performing a heating treatment on the antigen protein chip under the presence of a protein modifier and reducer; operating serum and an anti-fluorescent protein non-human antibody on the antigen protein chip after the heating treatment; reacting a Cy5 labeled anti-human Ig antibody and a Cy3 labeled anti-non-human Ig antibody sequentially through two stages; and calculating a fluorescence intensity ratio (Cy5/Cy3). <P>COPYRIGHT: (C)2012,JPO&INPIT |
