<P>PROBLEM TO BE SOLVED: To provide the manufacture and use of a diverse repertoire of functional heavy chain-only antibodies that undergo affinity maturation, the manufacture and the use of a diverse repertoire of class-specific heavy chain-only antibodies and the antibody heavy chain functionality, preferably the antibody heavy chain binding functionality, the manufacture and use of multivalent polypeptide complexes with effector activity in a constant region and an optionally selective additional effector functionality, a method for generating sufficiently functional heavy chain-only antibodies in transgenic mice in response to antigenic challenge, and in particular, a method for generating human antigen-specific high-affinity heavy chain-only antibodies of an optionally selected class or mixture of classes and the isolation and expression of fully functional VH antigen-binding domains. <P>SOLUTION: VH heavy chain-only antibodies expressing different kinds of VH heavy chain gene loci and making transgenic mammals with reduced capacity of producing light chain-containing endogenous antibodies immune with antibodies are manufactured. <P>COPYRIGHT: (C)2012,JPO&INPIT |
