The present invention provides attenuated invasive bacteria, which are useful as bacterial vectors for introducing peptide and/or nucleic acids into mammalian cells, tissue, organs or organisms,. In detail, the present invention provides bacteria comprising a transgene, for example containing an expression cassette for a structural peptide, wherein the expression cassette is under the control of a specific promoter element. According to the invention, the promoter element in the absence of the corresponding inductor substance essentially eliminates the activity of the expression cassette comprising the transgene, e.g. essentially eliminating transcription of nucleic acid sequences operably linked to the promoter element. According to the invention, the promoter element originates from operator regions of bacterial origin, which are regulatable by saccharides, for instance inducible by increasing concentrations of a saccharide. |
