TOLEROGENIC DENDRITIC CELLS OBTAINED WITH HIGH DOSAGES OF INTERFERON-GAMMA

During intensive studies leading to the submitted invention, local inventors found out that dendritic cells cultivated in the presence of high concentrations of IFN-gamma are acting tolerogenically and posses a reduced ability of induction of proliferation of allogenic CD4+ cells T in vitro, produce low quantities of IL-12p70, induce allogenic naive CD4 cells T to become IFN-gamma low IL-10 producing CD4+ cells T and inhibit the formation of IFN-gamma in activated allogenic CD4+ cells T. Thus, the submitted invention, in the first aspect, ensures the preparation of tolerogenic dendritic cells treated with high dosages (greater than 500 U/ml) of IFN-gamma. The submitted invention also ensures the application of tolerogenic dendritic cells induced by IFN- gamma for prevention and treatment of diseases or conditions in which the application of immunosuppressants is indicated. Such conditions represent especially autoimmune diseases, asthma and conditions after allogenic transplantation of cells, organs and tissues.
