<p>The method involves generating an illumination light beam (24) and selecting an illumination area of a sample (40). The illumination light beam is split into three partial beams (30). The partial beams are focused onto the illumination area using a microscope optical system of a fluorescence microscope (20) so as to excite fluorescent dye particles in the illumination area to fluoresce. Fluorescent light (44) emitted by the dye particles is detected. A diffusion coefficient representative of diffusibility of the respective fluorescent dye particles is determined based on the fluorescent light. An independent claim is also included for an apparatus for performing fluorescence correlation spectroscopy, comprising a fluorescence microscope and a fully-depleted silicon detector.</p> |
