In the present invention, there is disclosed a method of purifying capsular polysaccharides from cellular components of gram-negative and gram-positive bacteria, the method comprising: (a) extracting the capsular polysaccharide from cellular components by contacting the isolated bacterial cells, concentrated bacterial supernatants from homogenized bacterial cells or conditioned medium or pelleted cells with a base reagent to obtain a mixture wherein the pH of the mixture is between about 9 and 14, and (b) separating the extracted capsular polysaccharide from impurities resulting from the cellular components wherein said impurities comprise compounds being selected from the group consisting of proteins and nucleic acids. |
