<P>PROBLEM TO BE SOLVED: To provide a method for humanizing antibodies based on selection of variable region framework sequences derived from human antibody gene. <P>SOLUTION: The selection is based on comparing the canonical CDR (complementarity determining region) structure type of CDR in variable region of nonhuman antibodies with the canonical CDR structure type of corresponding CDR derived from the library of the human antibody sequence (preferably germ line antibody gene segment). The human antibody variable region having the canonical CDR structure type resembling to the nonhuman CDR forms a subset of member human antibody sequences for selecting human framework sequences. The members of these subsets can be further ranked by amino acid similarity between the human CDR sequence and the nonhuman CDR sequence. The human sequences with higher rank are selected to provide framework sequences for constructing chimera antibodies. <P>COPYRIGHT: (C)2011,JPO&INPIT |
