PURPOSE: A method for preparing biodegradable polymeric microspheres is provided to enhance drug effect without initial excessive release. CONSTITUTION: A biodegradable polymeric microsphere containing glucose controlling peptides constantly releases the glucose controlling peptides. The glucose controlling peptide is synthetic exedin-4, analogue, or agonist thereof. The biodegradable polymer is poly-L-lactic acid, poly-D-lactic acid-Co-glycolic acid, poly-L-lactic acid-Co-glycolic acid, or poly-D,L-lactic acid-Co-glycolic acid. A method for preparing the biodegradable polymeric microsphere comprises: a step of adding an organic solvent to polymers to obtain a polymer solution; a step of adding glucose controlling peptide to the polymer solution to obtain a drug dispersion; a step of adding alcohol or mixture of alcohol and organic acid to prepare a drug dispersion solution; and a step of preparing sphere.
