PROBLEM TO BE SOLVED: To provide a method for easily identifying fine distinction of base sequence. SOLUTION: There are disclosed a method for detecting a sequence mismatch between a sample single-stranded nucleic acid molecule and a standard single-stranded nucleic acid molecule, comprising a step in which a nucleic acid molecule which is a partially double-stranded nucleic acid molecule comprising a single-stranded nucleic acid molecule (a)complementary to the sample single-stranded nucleic acid molecule and one or two single-stranded nucleic acid molecules (b) which hybridize with part of the single-stranded nucleic acid molecule (a), the region which assumes a single-stranded structure in the partially double-stranded nucleic acid molecule being complementary to a region comprising a site where the sequence mismatch is anticipated in the sample single-stranded nucleic acid molecule, is brought into contact with the sample single-stranded nucleic acid molecule under conditions which allow hybridization, and a step of detecting the sequence mismatch characterized by comprising the presence or absence of the sequence mismatch utilizing the fact that the exchange rate is rapid when there is no sequence mismatch and the exchange rate is slow when there is a sequence mismatch.
