[Also in Division C3] 15N-L-Glutamic acid having an isotopic content of more than 90% is prepared by reacting 15N-L-aspartic acid and α-keto-glutaric acid in a neutral medium in the presence of pyridoxal phosphate and an aspartate aminotransferase enzyme. A weakly basic ion-exchange resin is preferably present in the reaction mixture and the mixture is normally allowed to become acid to permit decarboxylation of the oxaloacetic acid formed. The 15N-L-aspartic acid may itself be formed by reacting sodium fumarate and 15N-ammonium chloride dissolved in a neutral buffer in the presence of an aspartase enzyme preparation.
