An injection port assembly for a liquid column chromatograph 28 which is operated at high pressure comprises a supply inlet 24 feeding solvent up annular space 40 and down tube 46 into the column, the top of the port assembly being sealed during the operation of the column by closure member 48 (see Fig. 6) which comprises a stem portion 54 which is held by set screws 72, and part of which is threaded at 56. The bottom face of stem 54 bears against a backup disc 70 to a polytetrafluoroethylene disc 68 which is pressed into chamfer 44 when closure member 48 is screwed down tight on the port assembly, thereby forming a sealing bleb. When a sample is to be introduced, closure member 48 is removed and replaced by a syringe guideway assembly 74 (see Fig. 4), the upper end of which is shaped to receive the syringe. When in position, the syringe needle lies along passage 82, and is thereby aligned with passage 38 and tube 46, into which the sample is introduced. The syringe assembly is then removed and the high pressure closure member 48 replaced before separation is commenced. |
