PROBLEM TO BE SOLVED: To provide a method for isolating and purifying nucleic acid, which has excellent separation performance and improved cleaning efficiency, is simple and rapid, has excellent automation and miniaturization suitability and in which a porous membrane having substantially the same isolating performance and being mass produced is used. SOLUTION: The method of isolating and purifying a nucleic acid, comprises the steps of: (1a) passing a sample solution containing the nucleic acid through a nucleic acid-adsorptive porous membrane to adsorb the nucleic acid to the nucleic acid-adsorptive porous membrane under a centrifugal force condition; (2a) passing a wash solution through the nucleic acid-adsorptive porous membrane to wash the nucleic acid-adsorptive porous membrane while adsorbing the nucleic acid under a centrifugal force condition; and (3a) passing a recovered solution through the nucleic acid-adsorptive porous membrane to desorb the nucleic acid from the nucleic acid-adsorptive porous membrane under a centrifugal force condition. The other embodiment of the invention discloses the method of isolating and purifying a nucleic acid, comprising the steps of: (1b) passing a sample solution containing the nucleic acid through a nucleic acid-adsorptive porous membrane to adsorb the nucleic acid to the nucleic acid-adsorptive porous membrane under a reduced pressure condition; (2b) passing a wash solution through the nucleic acid-adsorptive porous membrane to wash the nucleic acid-adsorptive porous membrane while adsorbing the nucleic acid under a reduced pressure condition; and (3b) passing a recovered solution through the nucleic acid-adsorptive porous membrane to desorb the nucleic acid from the nucleic acid-adsorptive porous membrane under a reduced pressure condition or a centrifugal force condition.
