Provided is a method of producing functional stem cells or stem cell-like cells comprising culturing a non-human embryo or primary embryonal outgrowth from said embryo in the presence of a demethylation agent and isolation functional pluripotent cells. Further provided are corresponding stem cell isolation media comprising a demethylation agent.
