IMMUNOGLOBULIN FRAMEWORK DEMONSTRATING ENHANCED STABILITY IN INTRACELLULAR ENVIRONMENT AND METHOD FOR IDENTIFYING THE SAME

PROBLEM TO BE SOLVED: To provide a composition capable of being used as frameworks for creating very stable solubility single chain Fv antibody fragments.

SOLUTION: The frameworks are selected for intracellular performance, and are ideally suited to create scFv (single chain Fv fragment) antibody fragments or scFv antibody libraries for applications where stability and solubility are limiting factors for the performance of antibody fragments (for example, in the reducing environment of a cell). Such frameworks can be also used to identify highly conserved residues and consensus sequence which demonstrate enhanced solubility and stability.
