PROBLEM TO BE SOLVED: To provide a more inexpensive single nucleotide polymorphism (SNP) detection method free from SNP detection error in the predetermined SNP detection in a nucleic acid specimen by a mutant allele specific amplification method (MASA method). SOLUTION: The method includes the step of conducting PCR using a blocking agent consisting of an oligonucleotide that is preferentially hybridized with another base of SNP with which the primer is not hybridized, together with the primer specific to a specific base of SNP used in conventional MASA methods; wherein the blocking agent has ddNTP on the 3'-end.
