PROBLEM TO BE SOLVED: To provide a nucleic acid sequence containing unmethylated CpG dinucleotides that modulate an immune response including stimulating a Th1 pattern of immune activation, cytokine production, NK lytic activity and B cell proliferation. SOLUTION: An isolated nucleic acid sequence comprises at least one unmethylated CpG dinucleotide and has a formula: 5'N1X1CGX2N23' (wherein, CpG into which at least one nucleotide is continued is separated; X1is adenine, guanine or thymine; X2is cytosine or thymine; N may be any nucleotides, and N1+N2is about 0-26 bases with a proviso that N1and N2do not contain a CCGG tetramer or one or more CCG or a CGG trimer; and the nucleic acid sequence has about 8-30 base length).
