PROBLEM TO BE SOLVED: To provide a target substance detection element for detecting target substances with high sensitivity, and to provide a method of manufacturing the target substance detection element. SOLUTION: The target substance detection element includes: a linker layer provided at a part made of gold or silver on the surface of a metal structure; and a target substance capturing body bonded to the linker layer. The linker layer is made of nonassociated bodies of a chain polymer without any crosslinking structures. The polymer includes: a polymer comprising a repetition unit A having an amino group in a side chain and a repetition unit B having a carboxyl group in the side chain; and a polymer comprising a repetition unit C having an amino group and a carboxyl group in the side chain.
