PURPOSE: A buffer composition for promoting production of cacitriol or calcifediol is provided to reduce purification cost and to improve product quality. CONSTITUTION: A buffer composition for promoting cacitriol or calcifediol production contains: metal compound 0.01-0.3%(w/v) of FeCl_2, FeCl_3, FeSO_4, MnCl_2, or ZnSO_4; organic solvent 1-10% of acetone, dimethyl sulfuroxide (DMSO), ethanol, or methanol; cyclodextrin 0.1-5%(w/v); tris(hydroxymethyl)aminomethane0.01~1%(w/v); sodium succinate 0.01~1%(w/v); sodium chloride 0.01~1%(w/v); magnesium chloride 0.001~0.5%(w/v); and remainining amount of water.
