<P>PROBLEM TO BE SOLVED: To provide a caboxylation carrier for affinity chromatography capable of efficiently introducing a carboxylic acid into a spacer terminal under a condition more moderate than before using a small amount of a reagent, a method for manufacturing the same, a novel separating agent for affinity chromatography manufactured using the same, a method for manufacturing the same and the application of the separating agent. Ž<P>SOLUTION: The carboxylation carrier for affinity chromatography having a carboxylic acid group containing a thiol group and a ligand containing a primary amino acid are reacted with the surface of a carrier for chromatography using a dehydrocondensation agent to obtain the separating agent for affinity chromatography wherein the ligand containing the primary amino group is fixed to the carboxylation carrier by an amide bond.
