<P>PROBLEM TO BE SOLVED: To provide a DNA polymerase having improved fidelity in DNA replication, especially a DNA polymerase having thermal stability and improved fidelity in DNA replication. <P>SOLUTION: The DNA polymerase variant has enhanced 3'→5' exonuclease activity in cooperation with a DNA clamp and has an amino acid predetermined to be corresponding to arginine at 706 position of an amino acid sequence of Pyrococcus furiosus-derived wild type DNA polymerase, which is substituted with an amino acid except an amino acid having a positive charge by a homology analysis. <P>COPYRIGHT: (C)2010,JPO&INPIT |
