PROBLEM TO BE SOLVED: To measure mainly the thickness of a live cell, which has been impossible hitherto, though a measuring sample is not necessarily restricted to a cell, whereas a measuring means for the thickness of a live cell does not exist hitherto. SOLUTION: This method is constituted of: the first process wherein a measuring sample is disposed in liquid and measured, while changing a composition or a concentration of the liquid or both of them, and a liquid is specified, having such a condition of the composition, the concentration or the like that its refractive index is equal to the refractive index of the measuring sample; the second process wherein the refractive index of the liquid is measured by using a measuring reference body placed in the liquid specified in the first process, and the refractive index of the measuring sample is determined; and the third process wherein the measuring sample is disposed in liquid different from the first and second processes, and the height of the measuring sample is measured by using the refractive index of the liquid if the value thereof is known, or by using the refractive index of the measuring sample which is known by being measured previously by a similar method to the second process or by another method if the refractive index of the liquid is not known.
