<p><P>PROBLEM TO BE SOLVED: To increase the proportion of endoderm committed cells and pancreatic lineage cells in a culture of human embryonic stem cells which are undergoing differentiation. <P>SOLUTION: A method for producing a stem cell-derived cell population which does not have tumorigenic capability is provided. The method includes the following steps: (1) a step of culturing human stem cells into the form of embryoid bodies; (2) a step of suspending the cultured cells; and (3) a step of selecting the stem cells for lack of expression of stage-specific embryonic antigen-3/4 (SSEA-3/4), for the lack of the expression of stage-specific embryonic antigen-1 (SSEA-1), and selecting for the expression of epithelial cell adhesion molecule (EpCAM), and the resulting cell culture not forming teratomas when injected into immune-compromised mice. <P>COPYRIGHT: (C)2010,JPO&INPIT</p> |
