Disclosed is a biosensor comprising: a substrate, the substrate comprising a material having an optical axis (such as a birefringent substrate like PET); and a grating applied to the substrate, wherein the grating comprises features arranged in a plurality of parallel lines and wherein the lines of the grating are in substantial alignment with the optical axis of the substrate. Also disclosed is a method of manufacturing a biosensor, comprising the steps of: a) feeding a web of substrate material to a station, the web of substrate material having an optical axis; b) applying a grating to the substrate material at the station, wherein the grating comprises a plurality of parallel lines; wherein, in the performance of step (b), the grating is applied to the substrate in a manner whereby the lines of the grating are in substantial alignment with the optical axis of the web of substrate material.
