<P>PROBLEM TO BE SOLVED: To provide a method using chain-termination reaction to detect only methylated cytosine on a DNA chain. Ž<P>SOLUTION: The position of methylated cytosine in the target nucleic acid is detected by following steps: (i) only cytosine in the target nucleic acid having methylated cytosine is converted into uracil; (ii) uracil-containing nucleic acid is amplified by PCR; (iii) (a) reaction extending a chain to terminate to make ddCTP as the terminal end, by using primer, ddCTP and dNTP against complementary strand of a strand having methylated cytosine of amplified products or (b) reaction extending a chain to terminate to make ddCTP as the terminal end, by using primer, ddCTP and dNTP against complementary strand of a strand having no methylated cytosine of amplified products, is performed to PCR amplified product; and (iv) the nucleic acid fragments are divided to determine methylated cytosine part of the target nucleic acid.
